Osteosarcoma is the most common primary bone cancer. According to medical and biological studies, it has a high genetic complexity, thus, to differentiate the mechanisms of appearance and evolution of this disease is a difficult task. In this paper, we use three simplest and well known mathematical models to describe the behavior of several cell lines of osteosarcoma. First, we use a potential law to describe the tumor growth in immunosuppressed mice; with it we show that the variation of tumor (...) growth has a sublinear behavior without the blow-up phenomenon. Second, the logistic model is used to obtain a good aproximation to the rates of proliferation in cell confluency in in vitro experiments. Third, we use a linear reaction-diffusion model; with it, we describe the diffusion behavior for some cell lines. These three models allow us to give a classification of cell lines according to the rates of tumor growth and proliferation and to the diffusion coefficient. A relationship is found between the rates of the tumor growth, the diffusion coefficient and tumorigenicity. Experimental data are extracted from Lauvrak et al. Background: Nursing students on clinical placements as part of their professional training are routinely faced with situations involving ethical conflicts. The initial act of perceiving a situation as causing an ethical dilemma is the result of both the students’ personal values, drawn from their culture and families, and of the professional knowledge and values that they have acquired through training and experience. Objectives: Nursing students’ experiences on clinical placements in primary care settings were investigated in order to identify situations that (...) they perceived as involving ethical conflict and describe the elements they took into consideration during their decision-making processes in these situations. Methods: The research design was qualitative descriptive case study. Around 50 students from three different intakes to a nursing degree answered a questionnaire and discussed it in focus groups. Ethical considerations: The study was designed in accordance with the principles guiding research with human beings and was approved by the Human Research Ethics Committee. Results: Synthesised into two principal axes: (a) ethical conflicts in primary care, linked with the domains of working processes, professional nursing ethics and human and social rights and (b) students’ decision-making processes – realisation, reflection and intervention. Conclusion: The student nurses saw themselves both as actors and spectators in situations involving ethical problems and demanding moral deliberation, demonstrating the ability to base their arguments soundly. They tended to emphasise the possibilities offered by dialogue and that different ethical values must be respected to find fair solutions to ethical problems. (shrink)

Proximity and interpersonal contact are prominent components of social connection. Giving affective touch to others is fundamental for human bonding. This brief report presents preliminary results from a pilot study. It explores if exposure to bonding scenes impacts the activity of specific muscles related to physical interaction. Fingers flexion is a very important component when performing most actions of affectionate contact. We explored the visuo-motor affective interplay by priming participants with bonding scenes and assessing the electromyographic activity of the fingers (...) flexor muscle, in the absence of any overt movements. Photographs of dyads in social interaction and of the same dyads not interacting were employed. We examined the effects upon the electromyographical activity: during the passive exposure to pictures, and during picture offset and when expecting the signal to perform a fingers flexion task. Interacting dyads compared to matched non-interacting dyads increased electromyographic activity of the fingers flexor muscle in both contexts. Specific capture of visual bonding cues at the level of visual cortex had been described in the literature. Here we showed that the neural processing of visual bonding cues reaches the fingers flexor muscle. Besides, previous visualization of bonding cues enhanced background electromyographic activity during motor preparation to perform the fingers flexion task, which might reflect a sustained leakage of central motor activity downstream leading to increase in firing of the respective motor neurons. These data suggest, at the effector level, an implicit visuo-motor connection in which social interaction cues evoke intrinsic dispositions toward affectionate social behavior. (shrink)

The current study examined the effects of an intervention aimed at blocking the transfer of frustration from a previous experience (i.e. recall task) to a subsequent and unrelated task (i.e. ultimatum bargaining task). Participants who went through the intervention were more likely to accept unfair offers in the ultimatum bargaining task than those who did not go through the intervention. These results show that participants who were blocked from transferring their feelings of frustration from the recall task to the subsequent (...) bargaining task (no-transfer condition) more likely accepted unfair offers than those who inadvertently transferred their feelings of frustration (transfer condition). The effect of conditions on accept-reject decisions in the ultimatum bargaining was mediated by reported feelings of frustration. (shrink)

There is growing attention towards atypical brain-body interactions and interoceptive processes and their potential role in psychiatric conditions, including affective and anxiety disorders. This paper aims to synthesize recent developments in this field. We present emerging explanatory models and focus on brain-body coupling and modulations of the underlying neurocircuitry that support the concept of a continuum of affective disorders. Grounded in theoretical frameworks like peripheral theories of emotion and predictive processing, we propose that altered interoceptive processes might represent transdiagnostic mechanisms (...) that confer common vulnerability traits across multiple disorders. A deeper understanding of the interplay between bodily states and neural processing is essential for a holistic conceptualization of mental disorders. (shrink)

The coronavirus disease pandemic fundamentally disrupted humans’ social life and behavior. Public health measures may have inadvertently impacted how people care for each other. This study investigated prosocial behavior, its association well-being, and predictors of prosocial behavior during the first COVID-19 pandemic lockdown and sought to understand whether region-specific differences exist. Participants from eight regions clustering multiple countries around the world responded to a cross-sectional online-survey investigating the psychological consequences of the first upsurge of lockdowns in spring 2020. Prosocial behavior (...) was reported to occur frequently. Multiple regression analyses showed that prosocial behavior was associated with better well-being consistently across regions. With regard to predictors of prosocial behavior, high levels of perceived social support were most strongly associated with prosocial behavior, followed by high levels of perceived stress, positive affect and psychological flexibility. Sociodemographic and psychosocial predictors of prosocial behavior were similar across regions. (shrink)

All rights reserved.We present new observations of the low-mass companion to HD 984 taken with the Gemini Planet Imager as a part of the GPI Exoplanet Survey campaign. Images of HD 984 B were obtained in the J and H bands. Combined with archival epochs from 2012 and 2014, we fit the first orbit to the companion to find an 18 au orbit with a 68% confidence interval between 14 and 28 au, an eccentricity (...) of 0.18 with a 68% confidence interval between 0.05 and 0.47, and an inclination of 119°with a 68% confidence interval between 114°and 125°. To address the considerable spectral covariance in both spectra, we present a method of splitting the spectra into low and high frequencies to analyze the spectral structure at different spatial frequencies with the proper spectral noise correlation. Using the split spectra, we compare them to known spectral types using field brown dwarf and low-mass star spectra and find a best-fit match of a field gravity M6.5 ±1.5 spectral type with a corresponding temperature of K. Photometry of the companion yields a luminosity of log=2.88 ± 0.07 dex with DUSTY models. Mass estimates, again from DUSTY models, find an age-dependent mass of 34 ±1 to 95 ±4 M Jup. These results are consistent with previous measurements of the object. (shrink)

In this chapter, we recount some of the most pressing objections to character scepticism, pointing out their limitations and, when appropriate, incorporating their suggestions. From here, we consider what empirically informed moral improvement might look like by turning to the skill analogy. While the skill analogy provides a realistic rubric for becoming a better person, many of the questions concerning the details of how moral improvement might take place remain unanswered. When developing expertise in domains like chess and morality, a (...) wide range of factors will likely be important and it is unlikely than any one individual factor will be especially important. Given this, for any account of moral improvement, our optimism should be bounded: the effect of any particular intervention is likely to be limited, in both magnitude and domain. Lastly, we consider how character scepticism has reshaped the way we think about moral responsibility, whether this is cashed out in reasons-responsiveness or self-expression accounts. While both views face challenges, by distinguishing the possession of the responsibility-making feature from failures to manifest that property in behaviour, we gain a certain degree of wiggle room that allows us to accommodate empirical findings while holding on to notions of responsibility. This chapter highlights that the ‘person-situationism debate’ has expanded far beyond its beginnings, giving way to new accounts of moral improvement and moral responsibility. (shrink)

In ecology, if the considered area or space is large, the spatial distribution of individuals of a given plant species is never homogeneous; plants form different patches. The homogeneity change in space or in time (in particular, the related change-point problem) is an important research subject in mathematical statistics. In the paper, for a given data system along a straight line, two areas are considered, where the data of each area come from different discrete distributions, with unknown parameters. In the (...) paper a method is presented for the estimation of the distribution change-point between both areas and an estimate is given for the distributions separated by the obtained change-point. The solution of this problem will be based on the maximum likelihood method. Furthermore, based on an adaptation of the well-known bootstrap resampling, a method for the estimation of the so-called change-interval is also given. The latter approach is very general, since it not only applies in the case of the maximum-likelihood estimation of the change-point, but it can be also used starting from any other change-point estimation known in the ecological literature. The proposed model is validated against typical ecological situations, providing at the same time a verification of the applied algorithms. (shrink)

It is assumed that social bonds in humans have consequences for virtually all aspects of behavior. Social touch-based contact, particularly hand caressing, plays an important role in social bonding. Pre-programmed neural circuits likely support actions (or predispositions to act) towards caressing contacts. We searched for pre-set motor substrates towards caressing by exposing volunteers to bonding cues and having them gently stroke a very soft cloth, a caress-like movement. The bonding cues were pictures with interacting dyads and the control pictures presented (...) non-interacting dyads. We focused on the readiness potential, an electroencephalographic marker of motor preparation that precedes movement execution. The amplitude of the readiness potential preceding the grasping of pleasant emotional-laden stimuli was previously shown to be reduced compared with neutral ones. Fingers flexor electromyography measured action output. The rationale here is that stroking the soft cloth when previously exposed to bonding cues, a compatible context, would result in smaller amplitudes of readiness potentials, as compared to the context with no such cues. Exposure to the bonding pictures increased subjective feelings of sociability and decreased feelings of isolation. Participants who more frequently engage in mutual caress/groom a “significant other” in daily life initiated the motor preparation earlier, reinforcing the caress-like nature of the task. As hypothesized, readiness potentials preceding the caressing of the soft cloth were significantly reduced under exposure to bonding as compared to control pictures. Furthermore, an increased fingers flexor electromyographic activity was identified under exposure to the former as compared to the latter pictures. The facilitatory effects are likely due to the recruitment of pre-set cortical motor repertoires related to caress-like movements, emphasizing the distinctiveness of neural signatures for caress-like movements. We present a one per cent measurement of the cosmic distance scale from the detections of the baryon acoustic oscillations in the clustering of galaxies from the Baryon Oscillation Spectroscopic Survey, which is part of the Sloan Digital Sky Survey III. Our results come from the Data Release 11 sample, containing nearly one million galaxies and covering approximately 8500 square degrees and the redshift range 0.2 < z < 0.7. We also compare these results with those from the publicly released (...) DR9 and DR10 samples. Assuming a concordance Λ cold dark matter cosmological model, the DR11 sample covers a volume of 13 Gpc3 and is the largest region of the Universe ever surveyed at this density. We measure the correlation function and power spectrum, including density-field reconstruction of the BAO feature. The acoustic features are detected at a significance of over 7σ in both the correlation function and power spectrum. Fitting for the position of the acoustic features measures the distance relative to the sound horizon at the drag epoch, rd, which has a value of rd,fid = 149.28 Mpc in our fiducial cosmology. We find DV = at z = 0.32 and DV = at z = 0.57. At 1.0 per cent, this latter measure is the most precise distance constraint ever obtained from a galaxy survey. Separating the clustering along and transverse to the line of sight yields measurements at z = 0.57 of DA = and H =. Our measurements of the distance scale are in good agreement with previous BAO measurements and with the predictions from cosmic microwave background data for a spatially flat CDM model with a cosmological constant. © 2014 The Author Published by Oxford University Press on behalf of the Royal Astronomical Society. I don’t know whether undead beings exist. I also think it is an open question whether anyone is evil in, say, the way bad guys are depicted in supernatural horror films and serial killer movies. I do think it’s nevertheless puzzling that the undead are frequently portrayed as evil in that way. I’m inclined to think that if we were to stumble across any undead they would be less likely to be evil than any random live person we stumble across. (...) Consider this a call for some undead understanding. I am going to approach these conclusions in a roundabout way. First, I’ll try to sketch something of an ontology of the undead, an account of their nature and variety. A large part of the contemporary literature on dehumanization is committed to three ideas: (a) dehumanization involves some degree of denial of humanness, (b) such denial is to be comprehended in mental terms, and (c) whatever exact mechanisms underlie the denial of humanness, they belong in the realm of post-perceptual processing. This chapter examines (c) and argues that the awareness of minds might belong to perceptual processing. This paves the way for the possibility that dehumanization might, at least in part, (...) be a perceptual phenomenon, such that dehumanizers visually perceive the dehumanized as exhibiting lesser-than-human minds. It is perhaps unsurprising that the first systematic investigations of dehumanization approached the phenomenon as linked to contexts of war, genocide, extreme hatred, and violence. One guiding hypothesis was that dehumanizers exclude the dehumanized from a moral community of human beings, implicitly conceptualized as displaying distinct individualities and being embedded in caring interpersonal relations. By comprehending the dehumanized as deindividuated entities to which moral norms and considerations of fairness do not apply (Opotow 1990), dehumanizers are able to disengage from moral restrictions and self-sanctions (Bandura 1999). This paper develops the conjecture that the electromagnetic interaction is the manifestation of the torsion Ωμ of spacetime. This conjecture is made feasible by the natural separation of the connection ω μ v into “gravitational” and “electromagnetic” parts α μ v and β μ v , respectively, related to the metric and to the torsion. When α μ v is neglected in front of β μ v , the affine geodesics are shown to become the equations of motion of charged (...) particles with Lorentz force, for an appropriate choice of Ωμ. Since β μ v contains the factor q/m, neutral particles do not see the torsional part of the connection and behave as if Ωμ were zero, i.e., as in Einstein's theory of gravity (the same effect is obviously obtained for charged particles when β μ v 《 α μ v ).In addition to the factor q/m, the velocity of the test particle appears in Ωμ. This indicates that the appropriate context for this problem is to be found in velocity-dependent connections. The velocities are now coordinates and become the actual velocities of the test particles only in the system of equations that one solves for obtaining the affine geodesics in connections of this type.When written with differential forms, the combination of Maxwell's equations and of the pertinent form of the torsion suggests geometric field equations for electrodynamics. As for the gravitational part of the connection, it can be made to obey equations similar in form to the Einstein field equations. A unified geometric theory of electrodynamics and gravitation spontaneously emerges. The present state of the theory does not yet permit us to ascertain whether the right-hand side of the fully geometric, gravitational field equations corresponds to the energy-momentum tensor. In the process, we learn that the key element of Proclus’s theory is that Time, which orders events and distributes “lifetimes,” provides us with the possibility of understanding the world as an ordered, intelligible whole.The book contains four chapters providing a genealogy of sorts by systematically laying out Proclus’s inspiration from the philosophy of time in Plato, Aristotle, the Stoics, and Plotinus respectively. Chapter 1 starts from Plato’s Timaeus and the distinction between the “order of time” and the “flow of time.” The order of time is the intelligible, transcendent substance that causes the “flow of time,” that is, time as change. From the Republic, Proclus harvests “true Number” as determining the former, and the motions of the heavenly bodies dispensing lifespans to all beings as determining the latter. In Neoplatonic terms: Aristotle’s first intellect as first mover becomes the substantial Order of Time in Proclus, the transcendent monad, and his lower movers become the chain of emanated lower times as kinds of intelligence.In chapter 3, Stoic biology helps elaborate the understanding of time as the order within the cosmos, the world as a living being, and contributes the idea of one simultaneous present or “now” for everything. This cosmic life helps counter Aristotle’s criticism: All motions belong to specific substances but are part of the life of the cosmos. Vargas here also brings in an interesting link to more recent discussions in physics concerning the nature of simultaneity: How do we know whether things occurring far away from each other are simultaneous? The cosmic “now” and the notion of sympathy, which Proclus finds in synchronous processes, provide a metaphysical framework for such simultaneity.In chapter 4, Plotinus’s contribution to Proclus’s theory is discussed: the flow of time as the life of the world soul, which underlies all life in the cosmos and ties it together. It consists in the presence in the soul of the effects, that is, the external activity, of the emanated internal activity of Intelligence. Intelligence, according to Plotinus, must be alive because it is fully active, and its emanation in the world soul leads to the soul’s own external activity, that is, its uniform and discursive production of the world. This external activity is time in the strictest sense of the word. The main difference between Plotinus and Proclus is that for Proclus the flow of time is inferior to the eternal life of the world soul, belonging to its external activity, but for Plotinus it coincides with the world soul. Departing from modal logic, Jean-Yves Girard, as a logician interested in philosophy, presented a distinction between essentialism and existentialism in logic. Carlos Lobo reflected about the Girard’s concept to reinterpret the Husserlian Platonism in regard of the status of logical modalities. We start rescuing the notion of modal logic in the Edmund Husserl’s works, especially Formal and Transcendental Logic and First Philosophy. Developing this reflexion, we propose a new contribution to this discussion, reinterpreting the platonic influence in the Husserlian notions (...) of eidos and science, light of some readings of Lee Hardy and Johanna M. Tito. As a conclusion of this dialogue between Husserl and Girard, the method of eidetic variation is presented as a tool to review the idea of science, in a manner consistent with the phenomenological approach. The ‘Hart-miracle’, then the ‘Hart-phenomenon’ are analysed through surveying (1) the state of legal philosophising in England preceding Hart, (2) his professional career and (3) the early British reception of his work, including (4) the kinds of criticism as to its methodology and presuppositions as well as (5) its becoming a master type of jurisprudence with Oxford as a centre. Taking it as a mainstream, the continental tradition of encouragement to productive thinking characteristic even of Kelsenism is contrasted with its (...) worldwide reception, by the effect of channelling thinking. Its reception in Hungary with an overview of which trends of legal philosophising of the 20 th century has exerted or failed to exert an impact upon - as echoed by - theorising on law in Hungary is also considered. As concluded, the places in the imaginary Pantheon of 20 th -century legal philosophising occupied by Max Weber and Hans Kelsen (as accompanied by Eugen Ehrlich and Gustav Radbruch who are deemed to have been decisively significant minor masters), representing the first half of the century, are complemented indeed by the places occupied by H. L. A. Hart and Ronald M. Dworkin in representation of the second half of 20 th century. (shrink)

We deal with five aspects of the intrinsic connection of logic, ethics, and aesthetics in Wittgenstein's Tractatus: (a) the indication of what Wittgenstein called the "higher" of the world and language; (b) the conveyance of values through an intuition "sub specie aeternitatis"; (c) the discussion of internal properties of volitional totalities; (d) the reference to the metaphysical subject as the subject of volition (the will) (e) the indication of a "happy life". This article examines the commentary on Aristotle’s De anima by Alphonsus Vargas Toletanus, OESA († 1366). The commentary has been preserved in one manuscript, Cremona, Biblioteca Statale, Ms. 113 (Nl-12193), written in Bologna in 1475, and in at least five editions printed between 1477 and 1609. The article presents a detailed overview of the commentary by listing its quaestiones and conclusiones and by focusing on the sources used by Vargas. It argues that the general objective of the work (...) was to provide an overview of opinions on essential issues concerning the soul, to be used by students of the Augustinian Order. (shrink)

Este problema exige: primero, mostrar la forma en que una lógica de términos, la de Aristóteles, puede ser transformada en una la lógica de proposiciones y de esta manera automatizada su sintaxis; (...) y, segundo, lograr -en la automatización del razonamiento- la incorporación del sentido, es decir, de su semántica. Microsystems are described as contexts formed by a subject, their roles, their interactions, and a specific physical space and time, such as housing and the school environment. Although several studies suggest the importance of studying this type of environment and its repercussion on children’s development, in a Latin American context, few studies integrate the interaction of two primary settings in the development of executive functioning. The present study explores the effects of the quality of housing and school environments on the (...) perception of stress, decision making, and planning among children. A total of 114 children (43% girls and 57% boys, M age = 10.57) from a primary school located in a community classified as poor participated in the study. The following was measured: the environmental quality of classrooms, housing, stress, and executive functioning of children. The results reveal a model linking environmental quality levels in children’s homes and schools and executive functioning. We also obtain a mediating role of stress between microsystems and performance, finding a deficit in executive performance when children experienced higher levels of stress as a result of poor environmental quality both in their homes and in their schools. Aim: The presence of noise in urban environments is rarely considered a factor that causes damage to the environment. The primary generating source is transportation means, with vehicles being the ones that affect cities the most. Traffic noise has a particular influence on the quality of life of those who are exposed to it and can cause health alterations ranging from annoyance to cardiovascular diseases. This study aims to describe the relationship between the traffic noise level and the perceived annoyance (...) in the inhabitants of a city on the Northern Border of Mexico. The work carried out in a city represents the vulnerability characteristics: economic, social, and migratory of its sizable portion of the inhabitants. Due to that, it is impossible to identify precisely the number of residents as the number of vehicles in circulation.Methods: The streets and avenues with an annual average daily traffic of more than 1,000 vehicles were considered for the measurement of traffic noise. The equipment used was a vehicle gauge with non-invasive speed radar; type I integrating sound level meters, with their respective gauges and tripods. A questionnaire was applied to people living within 250 m of the streets and avenues in which the noise was measured.Results: The noise measurement found a parameter of LAeq estimated for 12 h during the day, exceeding 70 dBA. The data received from the questionnaire were statistically tested by using Pearson's correlation tests. A total number of 2,350 people were participated, of whom 1,378 were women and 972 were men. The age of participants is ranged from 18 to 75 years. The overall perception of traffic noise annoyance identified that 1,131 participants responded “Yes” as they considered the noise annoying. Participants who responded “No” as well as those who responded “Do not know” resulted in a total of 1,219 people.Conclusion: The results show that the population is desensitized to traffic noise and does not perceive it as an annoyance. The flow of vehicles and the type of vehicles are the significant factors for the propagation and increase in the traffic noise levels. Women present a considerable appreciation of traffic noise perception instead of younger people who demonstrate a higher tolerance to high-level exposure. Voluntades anticipadas: desafíos éticos en el cuidado del paciente Diretivas antecipadas de vontade: desafios éticos no cuidado do paciente Advance healthcare directives are intended for the individual to personally express their will and preferences about healthcare and treatment ahead of time. This literature review aims to describe the concept and structure of advance directives and the ethical aspects involved in patient care. Using the keywords “Advance Healthcare Directive” AND “Ethical Implication” and its Spanish equivalents, five databases were accessed: ProQuest, Philosophy, (...) PubMed, Web of Science and Scielo. Publications were narrowed down to the 2010-2018 period. Thirty-one articles were obtained and read critically. Results of this review were grouped into the following categories: concept, structure, clinical situations in which advance directives apply, strengths and limitations, as well as the ethical aspects involved. End of life-related experiences in which a person loses their ability to make decisions and cannot express their wishes, so it is impossible to know their will, are more common nowadays. Most health workers are not appropriately trained in the preparation and application of advance directives, which becomes an opportunity to research and delve deeper into the subject. Advance directives are a tool that gives health workers reliable information on a patient’s values and wishes, so it is vital to train them to provide respectful quality care. This paper is a critical discussion of Manuel Vargas’ Building Better Beings, focusing on the treatment of desert therein. By means of an analogy between morality and sport, I examine some seemingly peculiar implications of Vargas’ teleological and revisionary account of desert. I also consider some general questions of philosophical methodology provoked by revisionary approaches.

Introduction: Uncertainty is omnipresent in cancer care, including the ambiguity of diagnostic tests, efficacy and side effects of treatments, and/or patients' long-term prognosis. During second opinion consultations, uncertainty may be particularly tangible: doubts and uncertainty may drive patients to seek more information and request a second opinion, whereas the second opinion in turn may also affect patients' level of uncertainty. Providers are tasked to clearly discuss all of these uncertainties with patients who may feel overwhelmed by it. The aim of (...) this study was to explore how oncologists communicate about uncertainty during second opinion consultations in medical oncology.Methods: We performed a secondary qualitative analysis of audio-recorded consultations collected in a prospective study among cancer patients who sought a second opinion in medical oncology. We purposively selected 12 audio-recorded second opinion consultations. Any communication about uncertainty by the oncologist was double coded by two researchers and an inductive analytic approach was chosen to allow for novel insights to arise.Results: Seven approaches in which oncologists conveyed or addressed uncertainty were identified: specifying the degree of uncertainty, explaining reasons of uncertainty, providing personalized estimates of uncertainty to patients, downplaying or magnifying uncertainty, reducing or counterbalancing uncertainty, and providing support to facilitate patients in coping with uncertainty. Moreover, oncologists varied in their choice of words/language to convey uncertainty.Discussion: This study identified various approaches of how oncologists communicated uncertain issues during second opinion consultations. These different approaches could affect patients' perception of uncertainty, emotions provoked by it, and possibly even patients' behavior. For example, by minimizing uncertainty, oncologists may consciously steer patients toward specific medical decisions). Future research is needed to examine how these different ways of communicating about uncertainty affect patients. This could also facilitate a discussion about the desirability of certain communication strategies. SummaryIn the formative period of London's scientific instrument industry membership of a guild was a necessary step towards owning a business in the City. Through the guilds' formal system of apprenticeship, boys received first-class training in a skilled trade, and learned essential marketing and managerial techniques. By analysing the guilds' records of apprenticeship and subsequent guild life it is possible to determine chains of masters and apprentices by which the knowledge passed from generation to generation. At the same time, dates (...) can be established for the training and subsequent working life of many known makers. The chains of knowledge reveal new and often important masters who were previously unknown because, by chance, their instruments or advertisements have not survived to the present day. While outcomes for children with T-cell acute lymphoblastic leukemia have improved dramatically, survival rates for patients with relapsed/refractory disease remain dismal. Prior studies indicate that glucocorticoid resistance is more common than resistance to other chemotherapies at relapse. In addition, failure to clear peripheral blasts during a prednisone prophase correlates with an elevated risk of relapse in newly diagnosed patients. Here we show that intrinsic GC resistance is present at diagnosis in early thymic precursor T-ALLs as well as in a subset (...) of non-ETP T-ALLs. GC-resistant non-ETP T-ALLs are characterized by strong induction of JAK/STAT signaling in response to interleukin-7 stimulation. Removing IL7 or inhibiting JAK/STAT signaling sensitizes these T-ALLs, and a subset of ETP T-ALLs, to GCs. The combination of the GC dexamethasone and the JAK1/2 inhibitor ruxolitinib altered the balance between pro- and anti-apoptotic factors in samples with IL7-dependent GC resistance, but not in samples with IL7-independent GC resistance. Together, these data suggest that the addition of ruxolitinib or other inhibitors of IL7 receptor/JAK/STAT signaling may enhance the efficacy of GCs in a biologically defined subset of T-ALL.Leukemia advance online publication, 30 May 2017; doi:10.1038/leu.2017.136.
